Identification of Intestinal Microbial Community in Gallstone Patients with Metagenomic Next-Generation Sequencing

چکیده
Gallstone disease (GD) is one of the most common gastrointestinal diseases worldwide. Nowadays, intestinal microbiota are thought to play important roles in formation gallstones. In our study, human fecal samples were extracted for metagenomic next-generation sequencing (mNGS) on Illumina HiSeq platform, followed by bioinformatics analyses. Our results showed that there was a particular micro-ecosystem GD patients. contrast healthy people, sequences Bacteroidetes, Bacteroides and Thetaiotaomicron obviously more abundant patients at phylum, genus species levels, respectively. On other hand, glycan metabolism drug resistance, especially β-lactams, profound functions gut microbes compared those normal subjects. Furthermore, correlation analysis drew out existed significant relationship between serum levels biochemical indicators abundances illuminate both composition All all, study can broaden insight into potential mechanism how affect progression gallstones some extent, which may provide targets prevention, diagnosis or treatment GD.

Next generation sequencing data of a defined microbial mock community
Generating sequence data of a defined community composed of organisms with complete reference genomes is indispensable for the benchmarking of new genome sequence analysis methods, including assembly and binning tools. Moreover the validation of new sequencing library protocols and platforms to assess critical components such as sequencing errors and biases relies on such datasets. We here repo...
متن کاملMetagenomic sequencing of bile from gallstone patients to identify different microbial community patterns and novel biliary bacteria
Despite the high worldwide prevalence of gallstone disease, the role of the biliary microbiota in gallstone pathogenesis remains obscure. Next-generation sequencing offers advantages for systematically understanding the human microbiota; however, there have been few such investigations of the biliary microbiome. Here, we performed whole-metagenome shotgun (WMS) sequencing and 16S rRNA sequencin...
متن کاملMetagenomic Sequencing of an In Vitro-Simulated Microbial Community
BACKGROUND Microbial life dominates the earth, but many species are difficult or even impossible to study under laboratory conditions. Sequencing DNA directly from the environment, a technique commonly referred to as metagenomics, is an important tool for cataloging microbial life. This culture-independent approach involves collecting samples that include microbes in them, extracting DNA from t...
متن کاملViral Surveillance in Serum Samples From Patients With Acute Liver Failure By Metagenomic Next-Generation Sequencing
Background Twelve percent of all acute liver failure (ALF) cases are of unknown origin, often termed indeterminate. A previously unrecognized hepatotropic virus has been suspected as a potential etiologic agent. Methods We compared the performance of metagenomic next-generation sequencing (mNGS) with confirmatory nucleic acid testing (NAT) to routine clinical diagnostic testing in detection o...

W hen Antonie van Leeuwenhoek experimented with his microscopes in the late 17 century, he observed, among many other small things, bacteria from the mouth cavity. van Leeuwenhoek’s detailed descriptions of bacteria, spermatozoa, and cells marked the beginning of microbiology; his successors Robert Koch and Louis Pasteur stoked further interest in the microbial world with their discoveries of t...
